Genetic Analysis of Autosomal Recessive Cataract Gene, lr2, Using Intersubspecific F2 Mice

Abstract
To refine the position of lr2 gene prior to positional cloning, we have generated intersubspecific F©ü mice between cataract mutant mice, CXSD and wild genotype of Mus musculus molossinus (MSM1Ms or MSM). Total 456 progeny were obtained, of which genotypes were separated as wild homozygote : heterozygote : mutant homozygote to 100 : 226 : 130 in respect to D14Mit28 locus. We also determined the order of microsatellite markers using simple sequence length polymorphisms(SSLPs) and single strand conformational polymorphisms(SSCPs) analysis in the region between D14Mit102 and D14Mit33 adjacent to the lr2 locus. Total 20 mice showing recombination in the indicated region were obtained and the genotypes of the mice for 15 microsatellite markers were analyzed. The order and distance of microsatellite markers were following: (D14Mit102, 203) - 0.22 cM D14Mit143 - 0.11 cM - (D14Mit158, 262, 28, 87) - 0.11 cM - D14Mit86 - 0.22 cM - DI4Mit217 - 0.22 cM (D14Mit216, 39, 159, 237, 89) - 1.32 cM - D14Mit33.
